Hart Biologicals Protein C Clotting Kit is intended for the quantitative determination of plasma protein C activity by using a clotting end point assay.
Protein C is a vitamin K dependent plasma protein which shares homology with prothrombin and other vitamin K dependent coagulation factors. Protein C is a precursor of a serine which is activated by thrombin when thrombin is bound to a specific membrane protein, thrombomodulin. Activated protein C is a very specific protease which inactivates specifically the co-factors FVa and FVIIIa in a calcium and phospholipid manner.
While Protein C can be activated through thrombin, the rate of activation is slow. Under such conditions, protein C inhibitor protein inactivates protein C as fast as it is activated. The protein C clotting assay is performed in an analogous way as the assays for the individual coagulation factors. The sample is pre-diluted into a protein C deficient plasma which is prepared by immunoabsorption. Protein C is activated with Protac®, which is a rapid Protein C activator derived from Agkistrodon contortrix.
The inhibiting affect of Protein C is detected via the aPTT. The prolongation of the clotting time (caused by the inactivation of FVa and FVIIIa) is proportional to the activity of Protein C in the patient plasma.
